• Chromatographic Conditions
유기화합물 첨가제의 첨가와 함께 인산염 완충제는 이동상에 사용된다.
(1,2-propenol과 acetonitrile은 가장 자주 첨가제로 사용된다.)
Enantioselectivity과 머무름은 이동상화합물이 바뀜에 따라 조정될 수 있다.

• Storage Conditions
컬럼은 실온이나 그 이하의 온도에 두어야 한다. 짧은 기간의 보관(즉, 일주일)동안 10%의 2-propanol 수용액으로
컬럼을 채우기를 권장한다.
컬럼을 오랜시간 보관해야 할 때는 15%의 2-propanol 수용액으로 컬럼을 채우고, 냉장보관을 권장한다.

Purospher® HPLC columns are based upon a high-purity, metal free silica for excellent separations with very good peak symmetry.
The base material for Purospher® high-purity HPLC columns is made from tetraalkoxysilane.
Due to the absence of heavy metals in the silica matrix and in combination with a complete coverage of the silica surface, this stationary phase enables tailing-free chromatography of acidic, basic and chelating compounds. This is of particular advantage for method development.
The Purospher® product family comprises three different Purospher® HPLC packing materials:
► Purospher® RP-18
Purospher® RP-18 is polar endcapped and suitable for separations of strong basic or chelating compounds(no acidic compounds) and separations of hydrophilic compounds with a high percentage ofwater in the mobile phase.

Purospher® RP-18 endcapped is suitable for separations of complex samples with simple eluents.

LiChrospher® silica carrier are available with different modifications.
The polar modified phases LiChrospher® CN, LiChrospher® NH2 and LiChrospher® DIOL as well as LiChrospher® Si with no modification are best for normal phase HPLC.
Furthermore LiChrospher® PAH is highly efficient and selective for the separation of PAH; as well as LiChrospher® WP 300 RP-18 for the separation of peptides and low molecular weight proteins.
LiChrospher® packing materials are available as Hibar® RT column and as LiChroCART® cartridge in different lengths and different internal diameters (10 mm, 4 mm, 3 mm and 2 mm).
LiChroCART® 3 mm i.d. and 2 mm i.d. narrow bore cartridges for HPLC saves costs by reducing the solvent consumption and allows the handling of very small quantities while increased sensitivity as well as resolution are achieved.
LiChroCART® cartridges 4 mm i.d., 3 mm i.d. and 2 mm i.d. are compatible to the manuCART® "4" this facilitate faster and more flexible method adaptation to smaller bore columns.

The polar modified phases LiChrospher® CN, LiChrospher® NH2 and LiChrospher® DIOL as well as LiChrospher® Si with no modification are best for normal phase HPLC.
Furthermore LiChrospher® PAH is highly efficient and selective for the separation of PAH; as well as LiChrospher® WP 300 RP-18 for the separation of peptides and low molecular weight proteins.
LiChrospher® packing materials are available as Hibar® RT column and as LiChroCART® cartridge in different lengths and different internal diameters (10 mm, 4 mm, 3 mm and 2 mm).
LiChroCART® 3 mm i.d. and 2 mm i.d. narrow bore cartridges for HPLC saves costs by reducing the solvent consumption and allows the handling of very small quantities while increased sensitivity as well as resolution are achieved.
LiChroCART® cartridges 4 mm i.d., 3 mm i.d. and 2 mm i.d. are compatible to the manuCART® "4" this facilitate faster and more flexible method adaptation to smaller bore columns.
LiChroCART® cartridges 10 mm i.d. have to be used with manuCART "10"-II.

The COSMOSIL®5C18-MS-II column, taking advantage of a new end capping treatment can replace the COSMOSIL® 5C18-MS. The silica gel is end capped with polar groups for shielding effect, based on the chemical property of the packed silica gel to draw highly polar components from the mobile phase. As a result, the new end capping provides improved peak shape for basic compounds. The absorption of the silica gel is controlled because it does not interact with N atoms, therefore suitable for the separation of chelating compounds. Another advantage is that the 5C18-MS-II has similar retention characteristics to the 5C18-MS series: the HPLC conditions for the 5C18-MS are applicable to the newly developed column, 5C18-MS-II.

The durability under extremely high pH is greatly improved at the new COSMOSIL® 5C18-MS-II compared with current COSMOSIL® 5C18-MS. Though it is possible to use under alkaline condition for a limited time, after long exposure some deterioration may occur. For long use we recommend pH 7.5 and below.

Method: After the indicated length of incubation with the extremely high pH buffer, the column was washed and the effective plate number (N) for n-amylbenzene was measured.
The figure above shows the change in N as a function of incubation time. There is hardly any observed change in MS-II. On the other hand with MS when 10 hours or more passed rapid deterioration was observed.

► Characteristics
The COSMOSIL® 5C18-AR-II packed column features a polymeric and near-perfectly end capped type of C18 reversed phase material that employs an ultra-pure silica gel low in metal impurities. In addition, it has stronger acid resistance than the COSMOSIL® 5C18-AR. The COSMOSIL® 5C18-AR-II column is especially effective for the separation of chelating compounds as well as both acidic and basic compounds.
This series of columns is also highly recommended for a wide range of applications particularly in situations where monomeric-type column (for instance, COSMOSIL® 5C18-MS-II) is not as effective as expected.

The acid resistance of COSMOSIL® 5C18-AR-II is much improved compared with 5C18-AR. It retains high performance even in case of acidic mobile phase used to separate proteins and peptides.
